Agar oligosaccharides are obtained by the degradation of agar. Agar is a kind of natural polysaccharide colloid which is extracted from red algae seaweed (gracilaria, lily, etc.) and is abundant in the ocean. Agar oligosaccharides generally refer to oligosaccharides with a degree of polymerization of 2-10, including agar oligosaccharides (reducing end is inner ether galactose) and new agar oligosaccharides (reducing end is galactose). Agar oligosaccharide has the functions of enhancing immunity, anti-inflammatory and anti-oxidation. This experiment studies the ability of agar oligosaccharide to scavenge free radicals in vitro.
Agar oligosaccharide was prepared by acid hydrolysis method, and agar oligosaccharide A1 and agar oligosaccharide A2 were obtained. A1 is agarose oligosaccharide mainly composed of disaccharides and tetrasaccharides, and A2 is agarose oligosaccharide mainly composed of hexose and octanose. The inhibitory effects of agar-oligosaccharide A1 and agar-oligosaccharide A2 on O-2·and·OH free radicals and their scavenging effects on DPPH were determined by experiments. As a result, the agarose oligosaccharides A1 and A2 had a good inhibitory effect on O-2· free radicals, and the inhibitory effect increased with the increase of the sugar concentration. Compared with carnosine, the scavenging effect of A2 on O-2· is similar to that of carnosine, with IC50 of 0.54mg/ml; while the effect of A1 is lower than that of A2, with IC50 of 0.8mg/ml. Agarose oligosaccharides A1 and A2 have certain scavenging effects on •OH free radicals, and their inhibitory effects increase with the increase of sugar concentration. The scavenging effect of A2 on •OH was significantly higher than that of agarose oligosaccharide A1, with IC50 of 1.2mg/ml and 2.3mg/ml, respectively. The mass concentrations of agar oligosaccharides A1 and A2 to DPPH clearance rate of 50% were 4 mg/ml and 6.4 mg/ml, respectively. The test results show that agarose oligosaccharides A1 and A2 have good inhibitory effects on O-2· and •OH free radicals, and the inhibitory effect on O-2· free radicals is better than that on •OH free radicals. However, the scavenging ability of alcohol-soluble and fat-soluble DPPH free radicals is slightly poor. Tests have shown that agarose oligosaccharides A1 and A2 have a certain scavenging effect on O-2 , OH and DPPH free radicals, so they have good application value in anti-oxidation, and can be used to scavenge free radicals and prevent skin Prevention and treatment of diseases related to free radical oxidation such as aging, anti-radiation, and prevention of atherosclerosis.
